So many to chose from. It's hard not to go with Harry.

My mostly gun related would be.

I have always liked this one:

If I were a Glock lover I'd like this one:

And for the future we all look to:

I also like Jess V's from Predator "I ain't got time to bleed" (which is not necessarily gun related) but the less well known and even better response is as just before the explosion from his grenade launcher Jesse's partner says "You got time to duck?"
Well, certainly not my favorite, as most of you have already stated my absolute favorites, but here is one I often remember from Where Eagles Dare, where Richard Burton regains the edge over Colonel Turner, a German double agent holding a machine pistol at Burton:
-You're finished, colonel.
-You're overlooking something, aren't you?
-Not at all.
